摘 要:目的 探讨肠巨噬细胞及肠上皮细胞分泌肿瘤坏死因子 (TNF α)和一氧化氮 (NO)的规律及地塞米松、TNF α单克隆抗体及复方大承气汤的作用。 方法 体外培养肠巨噬细胞、肠上皮细胞及二者混合细胞 ,用脂多糖诱导 ,再经地塞米松、TNF α单克隆抗体及复方大承气汤分别处理。应用放免法检测TNF α ;用酶法检测NO。 结果 正常肠巨噬细胞可分泌少量的TNF α和一氧化氮 (NO)。脂多糖诱导组 ,这两种物质分泌显著增加 ,地塞米松、TNF α单克隆抗体及复方大承气汤处理组与脂多糖诱导组相比 ,这两种物质分泌显著降低。正常肠上皮细胞可分泌少量的TNF α和一氧化氮 (NO) ,但各组之间并无显著性差异。混合细胞结果与肠巨噬细胞相同。 结论 肠巨噬细胞和肠上皮细胞都是肠道屏障的重要功能细胞 ,肠巨噬细胞本身在内毒素刺激下分泌炎性介质TNF α和NO ,肠上皮细胞无明显分泌作用。地塞米松、TNF α单克隆抗体及复方大承气汤均可抑制肠巨噬细胞分泌TNF α和NO。提示在炎症反应状态下应用地塞米松、TNFObjective To observe the release of tumor necrosis factor alpha(TNF α) and nitric oxide(NO) in cultured isolated gut macrophages,enterocytes and mixed cells,and to evaluate the effects of dexamethasone(DEX),monoclonal antibody to tumor necrosis factor alpha(TNF-α MoAb)and Fufang Da Chengqi(复方大承气) decoction on the release of these cells through inflammatory mediators induced by LPS Methods The isolated gut macrophages,enterocytes and mixed cells were cultured.Each of the three kinds of cells was induced by LPS except control group,meanwhile,some parts of induced cells were treated separately by DEX,TNF-α MoAb and Fufang Da Chengqi(复方大承气) decoction.After being cultured for 6h,the supernatants were collected and frozen until TNF-α and NO were determined.Results The normal gut macrophages could release a little TNF-α and NO,produce significantly more after induced by LPS.When the gut macrophages were treated by DEX,TNF-α MoAb and Fufang Da Chengqi(复方大承气) decoction,TNF-α and NO levels significantly decreased as compared with the induced cells.The normal enterocytes could secrete a little TNF-α,DAO and NO,but there were no differences between the cells in the three groups.The mixed cells were similar to gut macrophages. Conclusion Serum TNF-α and NO will be increasing significantly when the gut barrier was damaged.It is suggested that the gut macrophages play important roles in this process.When induced by LPS,they can produce more TNF-α and NO.The enterocytes are not obviously concerned with the process.Each of DEX,TNF-α MoAb and Fufang Da Chengqi(复方大承气) decoction can suppress the induced gut macrophages producing TNF-α and NO,which show protective effect on the gut barrier.
