摘  要:目的用氧化低密度脂蛋白对培养的大鼠主动脉平滑肌细胞进行干预 ,观察其对血管平滑肌细胞表达MMP-2的影响。方法用组织贴块法体外培养大鼠主动脉平滑肌细胞 ,分组后分别予以0、25μg/mlLDL,25μg/mlOX -LDL进行干预。提取细胞总RNA和总蛋白分别进行RT-PCR ,westernblot ting检测 ,同时取出条件培养基经含1mg/ml明胶的10%聚丙烯酰胺凝胶垂直电泳行酶谱法(zymo_gra phy)检测分泌于培养基中的MMP-2活性。结果经mRNA、蛋白和酶活性检测 ,发现25μg/mlOX -LDL干预48h时 ,MMP-2的表达水平较对照和25μg/mlLDL明显增加。结论OX -LDL能促进血管平滑肌细胞表达MMP-2,这提示血管细胞外基质的降解在氧化脂蛋白诱发的动脉粥样硬化斑块产生和破裂机制中起着一定作用。Objective To investigate the effects of oxidized low density lipoprotein(OX-LDL)on expression of matrix metalloproteinase_2(MMP_2)in aortic smooth muscle cells of rats and the influence of oxˉidized lipoprotein on matrix remodeling in atherogenesis and plaque rapture.Methods The MMP-2mRNA was determined by reverse transcriptase_polymerase chain reaction(RT-PCR),MMP_2protein was measured by western blotting and MMP_2activity in conditioned medium was observed by gelatin zymography.Results In comparison with the control,there was no difference in the expression of MMP_2between control and25μg/ml LDL,whereas MMP_2activity in smooth muscle cells was significantly increased by25μg/ml OX_LDL(P<0.01).Conclusions OX_LDLcan enhance the expression and activity of MMP-2in smooth muscle cells,which may provide an explanation for the phenomenon that patients who have high concentration of LDL are prone to have atherosclerotic lesions and plaque rapture.
