摘  要:目的 :观察葛根素对氯化钾 (KCl)和L -谷氨酸 (L -Glu)引起的新生大鼠脑细胞钙超载的影响。方法 :应用Fura -2 /AM及双波长荧光分光光度计测定正常新生大鼠大脑皮层细胞和在加入KCl和L -Glu后大脑皮层细胞内钙离子浓度 ,并和经葛根素处理过的中药组作比较 ;西药对照组设为尼莫地平用药组。结果 :葛根素在终浓度 2 .4μmol /L时 ,对以KCl和L -Glu诱导的大脑皮层细胞内钙离子浓度的升高具有抑制作用 ;同对照组相比 ,在对以KCl诱导的大脑皮层细胞内钙离子浓度的升高的抑制作用两者无统计学意义 ;但对以L -Glu诱导的大脑皮层神经细胞内钙离子浓度升高的抑制作用 ,葛根素组要优于西药对照组。结论 :葛根素对KCl和L -Glu诱导的新生大鼠大脑皮层细胞内钙离子浓度的升高具有拮抗作用。Objective:To speculate the effect of puerarin(Pue),a chemical substance abstracted from a chinese herbal medicine-Gegen(Radix Puerariae),on free cytosolic calcium ion concentration(Ca 2+ )i overloading in cerebral cortex cells of neonatal rats.Methods:Fura-2/Am and double-wave-length fluorospectrophotometer are usedl to measure the(Ca 2+ )i in neurons;Testing the normal group of neurons of normal neonatal rats ,The excited group of neurons processed by KCl or L-Glu,the Pue group of neurons processed by Pue before adding KCl or L-Glu into the neuron suspension and the controlled group(Nimodipine,Nim)of neurons processed by Nim before adding KCl or L-Glu into the neuron suspension,Through the comparison among these groups,we assay the effect of Pue on(Ca 2+ )i in neurons with the stimulation of KCl and L-Glu.Results:At the concentration of 2.4 μmol/L,Pue can inhibit the increasing of (Ca 2+ )i in neurons induced by KCl or L-Glu.Comparing with control group,there is no difference between the effect of pue and Nim on inhibiting the increasing of (Ca 2+ )i induced by KCl in neurons;but the effect of Pue on inhibiting the increasing of(Ca 2+ )i induced by L-Glu in neurons is better than the effect of Nim.Conclusion:Pue has the effect of inhibiting the increasing of (Ca 2+ )i in neurons of neonatal rats induced by KCl or L-Glu.
