摘  要:目的 :探讨心肌细胞缺氧时黄芪的保护作用及其机制。方法 :将原代培养的乳鼠心肌细胞随机分为五组 :正常对照组 (N组 )、缺氧对照组(H组 )、缺氧加黄芪组 1(A1组 )、缺氧加黄芪组 2 (A 2组 )、缺氧加黄芪组3 (A3组 )。A 1组、A2组、A 3组于缺氧前加入黄芪 ,黄芪的终浓度分别为10mg/ml、10 0mg/ml、10 0 0mg/ml,缺氧 3 0min ,观察心肌细胞内超氧化物歧化酶 (SOD )活性和上清液中丙二醛 (MDA )、肌酸磷酸激酶 (CK )含量的变化。结果 :与正常心肌细胞相比 ,缺氧 3 0min ,培养上清液中MDA和CK浓度显著升高 (P <0 .0 1) ,且心肌细胞内SOD活性降低 (P <0 .0 1)。与H组比较 ,A 2组、A 3组提高SOD活性 ,MDA、CK水平降低 (P <0 .0 5 ) ,但是均未恢复正常 (P <0 .0 1~ 0 .0 5 )。结论 :一定浓度的黄芪可能通过抗氧自由基、稳定细胞膜来减少缺氧心肌细胞损伤 ,但该作用并非与浓度正相关。Objective: To investigate the protective effect of astragalus memberanaceus(AM) on the myocardium during anoxia and its mechanism.Methods:Primary cultured neonatal rat myocardial cell were randomly divided into five group:normal control group(N),hypoxic group(H),AM 1(A1),AM 2(A2),AM 3(A3).AM was added to A1,A2,A3 before hypoxia,and the terminal concentration was 10 mg/ml,100 mg/ml,1 000 mg/ml,respectively.After being hypoxia for 30 minutes,myocardium superoxide dismutas(SOD)activity,malondialdethyde(MDA) and creatine kinase(CK)concentrations in supernant were measured.Results:Compared with normal cell,MDA and CK content of hypoxic cell increased significantly(P<0.01),while SOD activity decreased,Compared with hypoxic cell(P<0.01),Compared with hypoxic cell,SOD activity elevated,while MDA,CK receded in A2,A3(P<0.05).However above indices didn ,t reach normal level(P<0.01~0.05).Conclusion:AM may be beneficial to hypoxic myocardium injury by antioxygen free radical,stablening cell membrance,but such effect didn ,t correlate to its concentration.
