摘  要:目的 观察电刺激对后肢缺血大鼠 V EGF 的调节。方法 采用 SD大鼠股浅动脉结扎法制备后肢缺血模型 ,应用RT-PCR和免疫组化方法检测电刺激对后肢缺血大鼠 VEGF m RNA和蛋白表达的影响。结果 刺激频率为 2 5 Hz,强度为 0 .1V不引起肌肉收缩。缺血模型建立 7d后连续电刺激 7d,VEGF m RNA的表达刺激组比非刺激组增加 3倍 ,VEGF蛋白增加两倍。结论 电刺激可促进后肢缺血大鼠 VEGF m RNA和蛋白的表达 。Objective To investigated the effects of electrical stimulation on the expression of vascular endothelial growth factor(VEGF)mRNA and protein in a rat model of hindlimb ischemia.Methods RT-PCR and immunehistology were used to detect the expession of VEGFmRNA and protein in tibialis anterior (TA) muscles.Results The frequency of stimulation was 25 Hz and strength was 0.1 V, which was far below the threshold for muscle contraction. After 7-day stimulation,there was a siginificant increase in blood flow within the muscle. VEGFmRNA was augmented 4-fold in stimulated TA muscles.Immuehistochemical analysis revealed that VEGF protein had 2-fold increase of unstimulated group.Conclusion Electrical stimulation conducted the nonmolecular therapeutic angiogenesis via VEGF expression.
