摘  要:目的 探讨长期应用吗啡的雌性大鼠骨密度 (BD)和骨代谢生化指标的变化及其与性激素的关系。方法 选用 3 0只3月龄SD雌性大鼠 ,随机分为 2组 :生理盐水对照组 (14只 )和盐酸吗啡用药组 (16只 )。用药组大鼠按剂量递增的原则皮下注射盐酸吗啡 12周 ,吗啡用量控制在治疗量和极量之间 ,从 3mg/kg逐渐增至 44mg/kg。对照组大鼠皮下注射同等体积的生理盐水 12周。两组大鼠于 12周末经股动脉放血处死。分别测定股骨远侧干骺端、股骨干、腰椎及全身BD ,血清Ca、ALP、BGP、TPAP和尿Ca、HOP的值。结果 吗啡用药组大鼠E2 、PROG、LH较对照组减少 (P <0 .0 5 ) ,FSH无显著变化 ;股骨远侧干骺端及腰椎BD较对照组降低 (P <0 .0 5 ) ,而股骨干及全身BD与对照组相似 (P >0 .0 5 ) ;血清ALP、BGP较对照组虽有增加 ,但无统计学差异 (P >0 .0 5 ) ,血清Ca、TPAP和尿Ca、HOP较对照组显著增加 (P <0 .0 5 ) ,且血清TPAP和尿HOP的变化与E2 呈负相关 (r =- 0 .43、- 0 .3 8)。结论 长期应用吗啡后大鼠骨形成和骨吸收这一偶连失衡 ,骨吸收的作用远远超过了骨形成的作用 ,骨吸收代谢增强的变化与E2 呈负相关。Objective To investigate the effect of long term administration of morphine on sex hormone, bone density and biochemical markers of bone metabolism in female rats and their correlation analysis. Methods Thirty female SD rats (3 months old) were randomly divided into 2 groups. The rats in long term exposure to morphine group were subcutaneously injected with gradually increasing doses of morphine (from 3 mg/kg to 44 mg/kg, 12 weeks), and the rats in control group were injected with saline. Rats in both groupswere sacrificed at the end of 12th week. Results The rats in morphine group showed lower E 2, PROG and LH levels. Serum tartrate resistant acid phosphatase (TPAP) and calcium, urinary hydroxyproline (HOP) and calcium were significantly higher in morphine group than in control group, and BD in lumbar vertebrateand distal femur in morphine group rats significantly decreased, compared with control rats. Conclusion After long term administration of morphine, bone absorption is markedly higher than bone formation in female rats, and biochemicalmarkers of bone absorption metabolism is negatively correlated with E 2, indicating that hormone replacement therapy could be used to decrease bone loss.
