牛膝中胡萝卜甙对成骨样细胞的促进增殖作用及其含量测定

摘　　要：目的 :考察牛膝中胡萝卜甙对成骨样细胞 UMR10 6的促进增殖作用及其含量测定方法。方法 :将从牛膝中提取出来的胡萝卜甙和成骨样细胞 UMR10 6共同体外培养 ,用 MTT法检测细胞增殖 ;用高效液相色谱法对胡萝卜甙进行含量测定 ,色谱条件为 :ODS固定相、水 -乙睛 (70∶ 30 )作流动相 ,检测波长 2 0 3nm、柱温 30℃、非那西汀为内标。结果 :牛膝中的胡萝卜甙对成骨样细胞 UMR10 6有较强的促进增殖的活性 ,3次实验的最高增殖率平均为 4 5 .8% ,HPL C方法回收率为 94 .2 % ,RSD为 3% ,在 4～4 0 μg/ m L范围内线性关系良好。结论 :确定了胡萝卜甙为牛膝中对成骨样细胞有促进增殖作用的活性成分之一 ,含量测定方法准确、快速 。Objective:To study the effect of daucosterol in Achyranthes bidentata BL(AB)on proliferation of UMR106 osteoblast like (OB like)cells and to determine its content in AB by HPLC method.Methods:Daucosterol isolated from AB was cultured with UMR106 OB like cells together in vitro and the proliferatioa of OB like cells determined by MTT assay.The chromatographic condition determining daucosterol included an ODS column(250mm×4.6mm,5μm),a mobil phase consisting of a mixture of water acetontrile(70∶30),detection wavelength of 203nm,and a column temperature of 30℃,Phenacetin was used as the interal standard.Results:The daucostero from AB had significant effect on Phenacetion of OB like cells,the proliferation was promoted by 45.8%(n=3).The average recovery of daucosterol was 94.2%(RSD=3.2%),the calibration was linear in the range of 4～40μg/mL(r=0.9988).Conclusion:Daucosterol was screened quickly by cultivating with OB like cells together in vitro;the HPLC method is accurate,fast and repeatable for the determination of daucosterol in AB.
