中药治疗对大鼠坐骨神经损伤后神经生长因子蛋白表达的影响

摘　　要：目的　研究党参、丹参、黄芪、生地等单味中药及复方神肌再生冲剂对大鼠坐骨神经损伤后神经生长因子 (nervegrowthfactor ,NGF)蛋白表达的影响。方法　雄性SD大鼠 10 8只 ,按手术先后随机分成党参、丹参、黄芪、生地、复方神肌再生冲剂和对照组 6组 ,每组 18只。按取材时间分为术后 2、4、8周 3个时间组 (每组 6只 )。显露大鼠右侧坐骨神经 ,于坐骨结节远端 0 .8cm处造成坐骨神经挤压伤。根据分组 ,术后每天给予不同的中药灌胃 ,对照组给予生理盐水灌胃。术后不同时间取紧邻挤压伤部位的远端坐骨神经干 ,应用免疫组织化学和图像分析的方法研究神经组织中NGF的表达并进行定量分析。结果　术后 4、8周组 ,丹参和复方神肌再生冲剂组 ,坐骨神经组织中的NGF蛋白高表达。结论　大鼠坐骨神经损伤后用中药丹参和复方神肌再生冲剂治疗 。Objective To determine the effect of Chinese single herbs including Dangseng, Dansheng, Huangqi, Shengdi and compound Chinese herbs on the expression of NGF protein after sciatic nerve injuries at rats. Methods One hundred and eight SD male rats were randomly divided into six groups: Dangsheng group, Dansheng group, Huangqi group, Shengdi group, compound herbs group and control group, with 18 in each. Each group was subdivided equally into 3 groups according to the different postoperative interval: 2,4,8weeks. The right sciatic nerve were exposed and crushed at the site 0.8 cm distal to sciatic notch. After surgery the rats were fed with different Chinese herbs at the experimental group and normal saline at the control group. At various interval after surgery, the sciatic nerve stump immediately distal to the crushed site was harvested. Immunohistochemistry and image analysis were done to evaluate the expression of NGF. Results The NGF expression in the sciatic nerve was higher in the Dansheng group and compound herbs group than that in control group at the fourth and eighth weeks after surgery. Conclusion Chinese herbs Dansheng and compound Chinese herbs can increase the expression of NGF protein after sciatic nerve injuries at rats.
