摘  要:在浓度为20%的PEG-8000介导的渗透胁迫下,沙丘芦苇的胚性悬浮培养物的生长受抑程度小于沼泽芦苇,相对细胞活力也较高,表现出比沼泽芦苇较高的耐渗透胁迫能力。对渗调物质含量的分析显示:两种生态型芦苇细胞中脯氨酸含量均随处理时间逐渐增高。沙丘芦苇的可溶性蛋白含量、蔗糖含量等表现出比沼泽芦苇更为显著的积累。两种生态型芦苇的渗透适应性细胞中均大量积累脯氨酸和糖类。说明有机渗透调节物质的积累对提高芦苇细胞抗渗透胁迫能力有显著贡献。While under osmotic stress mediated by 20% PEG8000,the suspension culture of dune reed showed less inhibition on growth and much viability than that of swamp reed.The analysis of osmolytes showed that the contents of free proline in both reed ecotypes increased along the time course and the contents of soluble protein and sucrose presented much significant accumulation in dune reed than in swamp reed.The acclimated suspension cultures of both reed ecotypes that grew in 20% PEG8000 for long term accumulated large amounts of free proline and sugar.These results suggested that the suspension culture of dune reed remained higher osmotic stress resistance than swamp reed and mainly through the accumulation of osmolytes.
