摘 要:目的:观察Ⅱ型胶原海绵修复兔膝关节软骨缺损的组织学变化。方法:将28只成年雄性新西兰兔制作膝关节缺损模型并随机分成2组,A组在缺损局部植入Ⅱ型胶原海绵,B组不植入以作对照。手术后2、4、6、8、10、12周分别作HE、Safranin O染色及S-100蛋白检测。结果:(1)HE染色显示A组2周后即出现新生软骨细胞,12周后新生软骨组织完全填满缺损区并与周围组织整合,而B组直至术后第12周仍由纤维组织所填充;(2)S-100蛋白免疫组化检测结果显示A组新生软骨细胞具有正常兔软骨细胞的表型;(3)Safranin O染色结果显示A组新生软骨细胞具有分泌软骨基质的功能。结论:Ⅱ型胶原具有较强的诱导软骨细胞生长的能力,其诱导生长的新生软骨具有正常透明软骨的表型和功能。Objective: To observe the histological change during the repair of local osteochondral defect in rabbits' knee by type II collagen. Methods: Twenty-eight adult male New Zealand white rabbits were used for preparing the model of cartilage defect in knee joint, and these rabbits were randomly divided into two groups: rabbits were treated by implanting type II collagen into keen joints with the articular cartilage defects (group A) and rabbits did not be treated as a control (group B). The tissue samples of rabbits' keen joints were collected and examined by the method of H & E, Safranin O and S-100 protein im-munostaining at 2,4,6,8,10 and 12 weeks after treatment.Results:(1)The H & E examination showed that the regeneration of articular cartilage were found in group A at 2-week timepoint.and the new cartilage tissue can completely fill all defect rgeions in group A after 12-week treatment ; while only fibrous tissues could be found in defect regions in group B at 12-week time-point. (2)The S-100 protein immunohistochemistry examination confirmed that the regenerated chondrocytes had normal chon-drocytic phenotype in the rabbits of group A. (3)The Safranin O examination also confirmed that the regenerated chondrocytes had a normal secreting function in the rabbits of group A. Conclusions:The type II collagen can improve the regeneration of articular cartilage,and the chondrocytes in regenerated cartilage tissue have normal chondrocytic phenotype and function.
