摘  要:万古霉素产生菌东方拟无枝酸菌 (Amycolatopsisorientalis) 5 13经紫外线 (3 0W ,2 5 3 7nm ,距离3 0cm ,照射 5 0s)诱变后 ,在含万古霉素 10 0 0 μg/ml的浓度梯度平板上筛选万古霉素抗性变株 ,得到一株万古霉素抗性变株A .orientalis 6 2 1,其摇瓶效价较出发菌株提高 2 3 % ,对万古霉素的抗性提高 2 0 0 %。传代试验表明该变株的高产性能遗传特性较稳定。用正交试验法对万古霉素发酵培养基进行了优化 ,与原发酵培养基相比 ,万古霉素的摇瓶发酵效价提高了 14 3 %。After the monospore suspension of Amycolatopsis orientalis 5 13 was treated by UV (30w, 253 7nm, 50s), the vancomycin resistant mutant of A.orientalis were isolated on the agar plates containing 1000μg/ml vancomycin. The vancomycin resistant mutants could produce more vancomycin than the parent strain and UV mutants. Among them, the productivity of A.orientalis mutant 6 21 was increased by 23% and its resistance were increased by 200% compared with the parent strain A.orientalis 5 13. Submerged culture indicated that the hereditary character of high productivity of A.orientalis 6 21 was stable. The fermentation medium of A.orientalis 6 21 was optimized by the orthonormal design method. Under the optimal medium condition, the productivity of A.orientalis 6 21 was raised by 14 3% further.
