摘 要:目的探讨将亲和吸附技术和不同的化学发光免疫分析技术相结合,进行甲胎蛋白(AFP)异质体测定的可行性。方法本实验室方法与Bayer Corporation,ARCHITECT i2000(Abbott Laboratories),Immulite(D.P.C.),ACCESS(Beckman Coulter Inc)和酶联免疫分析(CanAg Diag.AB)5种测定系统或试剂测定的比较,获得与本实验室检测AFP异质体上的差异。结果本实验室所采用的检测方法结果与上述5个测定系统或试剂测得的AFP异质体没有显著差异。结论亲和吸附法能将目前在临床实验室现用的AFP测定"升级"为AFP异质体的测定,这是一种适合临床诊断的方法,能在较短的时间内获得与现有方法相同或更好的结果,且有操作简便、结果可靠和测定成本低等优点。Objective To combined affinity adsorption with different chemiluminescence immunoassay systems for AFP variants determination , and evaluated the feasibility of clinical application. Methods Compared with results of AFP variants were obtained the five kinds of methods:Bayer Corporation,ARCHITECT i2000 (Abbott Laboratories),Immulite (D.P.C. ) , ACCESS (Beckman Coulter Inc) and ELISA (CanAg Diag.AB). Results There were no significant differences among these methods for AFP variants determination. Conclusion Affinity adsorption could upgrade any routine AFP assay to the AFP variants assay. It is a suitable clinical laboratory method of AFP variants assay. It may be give the same perhaps better result in a short time compared with the current methods, and has the advantages of easier handling, reliable result and lower costs.
