α-海藻糖对自制人血清生化质控品的保护作用

摘　　要：目的研究液态冷藏条件(4±1℃)下,海藻糖保护剂对ALT、AST、GGT、LDH、AFU等酶活性及其他生化指标的保护作用,并探讨海藻糖对酶等的最佳保护浓度,以提供一种人血清基质生化质控品的良好稳定剂。方法用惰性分离胶管(SSTⅡ管)收集混合血清,加入保护剂;采用酶动力学方法分别在当天、第1、2、3、4、5、6、7、8、9、10个月分别测定五种酶活力及其他生化指标,并计算酶活力及其他生化指标保护率(%)。结果在10个月时,0.20 mol/L海藻糖保护组的11种血清酶的平均保活率为92.60%,而无海藻糖保护组的11种血清酶的平均保活率为69.71%,海藻糖组的保活率是无海藻糖组的1.32倍, 海藻糖对酶等的最佳保护浓度为0.20 mol/L。结论通过实验研究证实了海藻糖对人血清基质生化质控品液态冷藏状态下具有保护作用,是一种多酶等复合质控品的良好稳定剂。Objective To explore the protective effect of α-trehalose on the activity and stability of enzymes such as ALT, AST, GGT, LDH and AFU in serum; Also to discuss the optimal protective concentration of α-trehalose for those enzymes in order to provide a reliable quality-control serum. Methods Collect mixed serum with SSTⅡ tube, add protective additive, measure the activity of blood samples on the same day, and then on the first, second, third, fourth, fifth, sixth, seventh, ninth, and tenth month, respectively, the variation for the activity and stability of the enzymes was also observed. Results In the tenth month, the average survival rate of the eleven serum enzymes of trehalose protection group is 2. 60% , 1. 32 times as large as that of the non-trehalose group. The optimal trehalose protective concentration is 0. 20 mol/L at 4℃. Conclusion Laboratory investigative evidence proves that the trehalose have protective effect on enzymes in liquid state under as lower as degree 4℃ refrigerator, and is suitable for popularization in clinical application.
