摘　　要：目的 :利用报告系统pEGFP观察pEGFP_N1_mSemcap2重组质粒肌肉注射小鼠后mSemcap2在体内的分布情况 ,以及对集合淋巴结 (PP结 )淋巴细胞表型的影响。方法 :将pEGFP_N1_mSemcap2重组质粒肌注小鼠后分别在第5 ,7天收集各组织细胞 ,利用FACS检测其在组织中的表达 ;取PP结淋巴细胞进行流式细胞术分析。结果 :重组质粒肌注小鼠后在不同时间点从小肠、肺脏、肾脏、肝脏、PP结、肠系膜淋巴结、腹股沟淋巴结等部位检测到了蛋白表达 ;小鼠PP结B2 2 0 + 细胞比例增高。结论 :肌肉注射重组质粒后mSemcap2在体内的组织分布与其天然分布不尽相同 ;Objective:To observe the distribution of mSemcap2 in the mouse body and its effect on lymphocytes of mouse Peyer′s patch following intramuscular(im) injection of pEGFP_N1_mSemcap2.Methods:Cells of various tissues were collected on days 5 and 7 after im injection of pEGFP_N1_mSemcap2.FACS was used to observe the distribution of mSemcap2 and the changes in surface markers of lymphocytes of Peyer′s patch. Results:The expression of mSemcap2 was detected in intestine, lung, kidney, liver, Peyer′s patch,mesenteric lymph node and inguinal lymph node at different time points following im injection of recombinant vector.The proportion of B220 + lymphocytes in the Peyer′s patch was increased. Conclusions:The distribution of mSemcap2 in the mouse body after im injection of pEGFP_N1_mSemcap2 was not totally coincident with the natural condition; mSemcap2 could change the phenotype of the lymphocytes of Peyer′s patch.
