摘 要:目的 :测定用Edman降解方法不能测序的结构特殊多肽的序列。方法 :用纳升电喷雾四极杆_飞行时间串联质谱的denovo测序方法进行测序。结果 :测定了亮丙瑞林、胸腺五肽及EPD等的多肽序列 ,这些多肽用Edman降解方法未能测定出序列。结论 :串联质谱测序法是Edman降解测序法的最好补充 ,且具有灵敏、快速和样品用量少的优点。Objective:To analyze the sequence of some special peptides which can not be sequenced by Edman degradation method.Methods and Results:Peptides were sequenced by de novo sequencing method of nano_ESI_MS/MS.Leuprorelin, thymopoietin and EPD were sequenced by this method, which had not been sequenced by Edman degradation method because of their special amino acid modifications or compositions.Conclusion:Nano_ESI_Q_TOF_MS/MS is the best complementary method to Edman degradation method in peptide sequencing because this method has no restriction to N_terminal modification and has high sensitivity, high mass accuracy and high speed.
