摘 要:采用随机扩增多态性DNA(RAPD)技术对南四湖的泥鳅 (Misgurnusanguillicaudatus)和大鳞副泥鳅(Paramisgurnusdabryanus)进行了初步分析。共用 10个 10bp的OPG组寡核苷酸片段作为引物 ,筛选出结果好的 5个引物 ,从泥鳅 4个体基因组DNA中扩增出 142个片段 ,共有片段 12个 ,特有片段 5个 ;从大鳞副泥鳅 3个体基因组DNA中扩增出 10 1个片段 ,共有片段 18个 ,特有片段 5个 ,在总扩增出的 2 43个片段中 ,共有片段仅 4个。所有扩增片段大小介于 0 2~ 3kb之间。用PoPGen32软件计算出个体间的遗传距离 ,依此进行UPGMA聚类得到的分子系统树与形态分类学的结果一致 ,这 7个体被聚成 2大类 ,正对应着形态学的 2个种 :泥鳅和大鳞副泥鳅。The genetic differentiation of Misgurnus anguillicaudatus and Paramisgurnus dabryanus from Nansi Lake,Shandong Province was studied by RAPD.Totally 243 amplified DNA fragments (200~3000bp) were obtained by using 5 random oligonucleotide primers from 7 samples of the two kinds of fish.Of them 4 fragments were shared by the 7 samples,18 fragments by 3 samples of Paramisgurnus dabryanus and 12 fragments by 4 samples of Misgurnus anguillicaudatus. RAPD phenogeam was constructed by UPGMA using genetic distance of samples calculated by PoPGen32 software.It has been suggested that the 7 samples were divided into 2 clusters,which is consistent with the results based on morphological study in which they belong to Misgurnus anguillicaudatus and Paramisgurnus dabryanus.
