摘 要:目的 :比较小鼠小肠上皮内淋巴细胞 (intestineintraepitheliallymphocytes ,iIEL)与脾脏T淋巴细胞的差异并寻找刺激iIEL活化的特异性抗原。方法 :分别提取新鲜分离的iIEL和脾脏T细胞的细胞膜蛋白 ,通过SDS PAGE和银染法比较二者的区别 ;用蛋白定量和磷定量方法 ,比较两种细胞的膜蛋白与膜磷脂比。提取Daudi细胞和小鼠小肠上皮细胞的细胞膜脂 ,将其制成脂质体 ,采用H3 TdR掺入法检测其刺激iIEL和脾脏T细胞增殖的能力。结果 :两种细胞的膜蛋白存在着质和量的差异。iIEL膜蛋白与膜磷脂之比高于脾脏T细胞。脾脏T细胞对Daudi细胞膜脂和小肠上皮细胞膜脂的刺激无反应 ,甚至有负反应 ;而iIEL对小肠上皮细胞膜脂刺激的增殖反应强于对Daudi细胞膜脂的。结论 :iIEL与脾脏T淋巴细胞在形态、表型、起源、发育以及功能上的差异存在相应的分子基础。小肠上皮细胞膜脂可能作为一种特异性抗原参与iIEL的活化。Objective:To compare the differences between mouse intestine intraepithelial lymphocytes(iIELs)and splenic T cells and search for the special antigen activating iIELs Methods:Had extracted cell membrane from iIELs and splenic T cells those were isolated freshly, and compared these two kinds of membrane proteins through SDS PAGE and silver staining Through quantitative analyses of protein and lipid of the two kinds of cell membranes,analyzed the ratios of protein to lipid of them In addition,prepared liposomes with membrane lipid from Daudi cells and mouse intestinal epithelial cells(IECs), and then added them to freshly isolated iIELs and splenic T cells respectively The proliferation of them was studied by the incorporation of 3H methyl thymidine Results:Had found that the membrane proteins of iIELs and splenic T cells were different qualitatively and quantitatively, and the ratio of protein to lipid of iIELs were higher than that of splentic T cells In proliferation experiment,had found that splenic T cells showed poor proliferation responding to the membrane lipid of Daudi cells and IECs However, iIELs can be activated by both of them and the effect of later one was stronger Conclusion:The differences between iIELs and splenic T cells in morphology, phenotypes, ontogeny and functions have the corresponding molecular bases The membrane lipid of IECs may be the special antigen that could activate iIELs
