动物心肌缺血中医证型规范化标准化研究(二)

摘　　要：为建立心肌缺血心气阴两虚证的标准化动物模型。以小站台水环境法剥夺快速眼动期(REM)睡眠 96小时 ,结合运用垂体后叶素制造大鼠心肌缺血气阴两虚证 ,分别给予丹参滴丸、半夏薤白栝蒌合剂、生脉饮、苏合香丸、左归丸、右归丸六种临床常见证型代表方进行治疗。 7天后分别测定心电图 ,左室功能 ,及心脏cAMP、cGMP、一氧化氮 (NO)含量和一氧化氮合酶 (NOS)活性。结果 :联合使用两种造模因子 ,可使大鼠心电图产生缺血性改变 (P <0 0 5 ) ,产生左室舒缩功能降低 ,(P <0 0 5 ) ,cGMP、NO含量明显升高 (P <0 0 5 )。辨证治疗组 (生脉饮组 )动物心电图和心功能指标明显改善 (与模型组比较P <0 0 5 )。结论 :联合运用睡眠剥夺和垂体后叶素可以制造大鼠心肌缺血气阴两虚证模型 。To establish a standard animal model of myocardial ischemia of heart qi yin deficiency syndrome,the rats were deprived of REM sleep for 96 hours and then given injection of pituitrin to cause myocardial ischemia of qi yin deficiency.They were treated respectively by'Danshen Drop Pill','Banxia Xiebai Hualou Mixture','Shengmai Decoction','Suhexiang Pill','Zuogui Pill'and 'Yougui Pill'.After 7 day treatment,the electrocardiogram,left ventricular function,the contents of cAMP,cGMP and NO in the heart as well as the activity of NOS were detected.Results:After modeling,the electrocardiogram showed myocardial ischemia( P <0 05),diminished systole and diastole of the left ventricle( P <0 05),and increased contents of cGMP and NO( P <0 05).In those rats treated by'Shengmai Decoction',the electrocardiogram and heart functions improved more obviously in comparison with those non treatment rats, P <0 05.It is revealed that deprivation of REM sleep and injection of pituitrin can make rats suffer from myocardial ischemia of qi yin deficiency,in which'Shengmai Decoction'can improve the electrocardiogram and heart functions.
