摘 要:通过木兰科29个种的trnL基因内含子的525个碱基序列及21个种的trnL-trnF基因间隔区的370个碱基序列测定和分析,构建的严格一致性材建议:①含笑属为相对一致的单系类群;②玉兰亚属的几个种形成一个单系类群,与木兰亚属的几个种无共衍位点;③木莲属为一个相对保守的类群,种间碱基差异很小。碱基位点差异分析表明:含笑和野含笑的trnL基因序列具2个位点的差异,结合形态学特征考虑,不支持David把两个种合并的处理;杂交鹅掌揪的trnL-trnF序列相对鹅掌楸有3个位点的变异和一个简单重复序列的插入;鹅掌楸的trnL-trnF序列与GenBank(AF040679)中已注册的序列有3个位点的差异。两个严格一致性树的CI值高达0.978和0.913,说明trnL内含子和trnL-trnF间隔区序列的平行演化在木兰科这几个属间发生频率非常低,只适于木兰科高等级的(属以上)的系统研究。The sequences of segments of the trnL intron from 29 species and trnL-trnF IGS from 21 species of Magnoliaceae were analyzed. The results from the strict consensus trees were consistent with the former systems: ①several species within the genera Michelia were homologious; ②the subgenera Yulania was extremely homologious and much far from subgenera Magnolia;③the species within Manglietia were highly conservative. The variation of sequence bases suggested:since the trnL intron of Mi. figo had two different bases from that of Mi. skinneriana ,it could not be combined with Mi. skinneriana;the trnL-trnF IGS of L. Chinese X L. tulipifera had 3 different bases and 4 bases insertion from that of Liriodendron chinense; the trnL-trnF IGS of L. chinense,which was sequenced,had 3 different bases in comparison with that of the report from GenBank(AF040679). The high consistency index implied that homoplasy was very low in trnL intron and trnL-trnF IGS of Magnoliaceae. The results indicated that these sequences could be used for the classification of Magnoliaceae at higher hierachy above genus.
