摘  要:目的 观察褪黑素 (melatonin ,MLT)对小鼠移植性肝癌生长的抑制作用及对小鼠腹腔巨噬细胞分泌的细胞因子的影响。方法 建立肝癌小鼠模型 ,连续 2周每日皮下注射MLT ,绘制肿瘤生长曲线并采用放射免疫法测定小鼠腹腔巨噬细胞分泌的IL 1β和TNF α的水平。结果 注射MLT的小鼠肿瘤生长速度明显 <阴性对照组 (P <0 .0 5 ) ,腹腔巨噬细胞IL 1β的诱生水平明显提高 (P <0 .0 5 ) ,但TNF α的水平却明显下降。结论 MLT能显著抑制小鼠移植性肝癌的生长 ,它对荷瘤小鼠的巨噬细胞功能具有选择性调节作用。Objective To observe the inhibitory effect of melatonin on the development of the transplanted hepatoma and to assess the regulation ability of melatonin on the cytokine production by peritoneal macrophages. Methods (1) Established the animal model by transplanting H22 hepatoma cells to Balb /c mice, then the mice received 14 consecutive daily subcutaneous injections of melatonin and mice in control group received normal saline. (2) Drew growth curve of tumor growth (3) After lipopolysaccharide (LPS, 10 /ml) treatment peritoneal macrophages isolated from mice after the mice received 5 days injection of melatonin the concentration of interleukin-1beta (IL-1β) and tumor necrosis factor (TNF-α) were tested in culture medium. Results (1) Growth of tumor was much slower in the mice received melatonin injection. (2) Melatonin stimulated IL-1β secretion ( P <0.05), but inhibited TNF-α secretion of peritoneal macrophages. Conclusion Melatonin could inhibit the development of transplanted hepatoma, and it has the ability to regulate cytokine production of peritoneal macrophages.
