摘 要:为了研究细胞活化后组蛋白磷酸化程度的改变以及对其免疫原性的影响。将正常淋巴细胞组蛋白人为加磷酸、活性组蛋白人为去磷酸 ,用Westernblot法检测其磷酸化程度 ;然后用以上不同组蛋白同系免疫BALB/c小鼠 ,用ELISA法测定IgG类抗组蛋白抗体、抗dsDNA抗体 ,用免疫荧光法检测抗核抗体核型及肾脏病理 ,用免疫印迹法测定可溶性核抗原抗体。结果发现淋巴细胞活化后 ,其组蛋白H1成分发生明显的丝氨酸磷酸化 ,对正常组蛋白人为加磷酸仍无免疫原性 ,活性组蛋白人为去磷酸后仅影响抗体产生的滴度 ,仍具有免疫原性。所以丝氨酸磷酸化在组蛋白的免疫原性改变方面起一定的作用 。To investigate the alteration of the histone phosphorylation level when lymphocytes were activated and whethr it has effects on its immunogenicity.The normal histone was artificially phosphorylated,while the active histone dephosphorylated,and the degree of phosphorylation was checked by Western blot analysis Syngeneic mice were immunized with these different histones,we detected the autoantibodies in serum,including anti dsDNA, anti histone, and anti ENA antibody The experimental results showed that H 1 component of histone was serine phosphorylated following lymphocyte activation There was no any effect of the artificial phosphorylation of the normal histone on its immunogenicity,while artificial phosphorylation of the active histone only influence upon the titers of antibody production,yet,it still persisted its immunogenicity These results suggest that serine phosphorylation may play certain roles in the immunogenicity of histone,but this is not the main factor
