摘  要:利用正交试验研究了培养基、2,4-D、蔗糖、脯氨酸、水解酪蛋白(CH)5个因素及组合对玉米自交系4112和P1幼胚愈伤组织诱导的影响,探讨了正交设计应用于玉米培养基筛选的可能性。结果表明:1)对4112诱导胚性愈伤的最优组合是8114培养基、2,4-D4mg/L、蔗糖80g/L、CH500mg/L,对P1最优组合是蔗糖N6培养基、2,4-D1mg/L、蔗糖20g/L、脯氨酸1400mg/L、CH500mg/L;2)不同玉米自交系的幼胚需要不同的糖浓度和培养基;3)适宜的2,4-D浓度可提高愈伤组织的诱导率;4)脯氨酸和水解酪蛋白对愈伤组织的形成有一定作用。The difference of callus initiation of maize inbred line4112and P1from young embryos on different media were compared.Experiments were conducted by using the orthogonal experiment design.The type of media,2,4-D,sucrose,proline and CH were used to compare their effects on callus initiation from young embryos.The pos-sibility of application of the orthogonal experiment method in tissue culture of maize inbred line was discussed.Re-sults showed that(1)the best combination for4112was8114culture media,2,4-D4mg /L,sucrose80g /L,CH500mg /L,for P1was N6media,2,4-D1mg /L?sucrose20g /L?proline1400mg /L,CH500mg /L;(2)the embryos of different maize inbred line species demanded different concentration of sucrose for callus initiation;(3)the suitable concentration of2,4-D was favorable for promoting the yield of callus;(4)it was quite useful for callus initiation to use proline and CH.
