摘　　要：目的　建立Ⅱ型胶原酶联免疫测定方法学 ,为临床骨性关节炎 (OA)的诊断及预后评价提供灵敏的检测手段。方法　建立Ⅱ型胶原酶联免疫夹心测定法 ,在动物实验的基础上对 2 4例正常人及 65例OA患者进行分组血清学检测 ,并与蛋白多糖的血清学检测进行比较。结果　Ⅱ型胶原与蛋白多糖高度相关 ,OA患者血清中两种指标明显高于正常人 (P <0 0 1,P <0 0 5 ) ;X光片显示OA患者B组、C组Ⅱ型胶原明显增高 (P <0 0 5 ) ;截骨术后半年及 1年Ⅱ型胶原明显降低 (P <0 0 5 )。Objective To develop the enzyme linked immunosorbent assay (ELISA) of type Ⅱ collagens and study clinical significance of type Ⅱ collagens in patients with osteoarthritis(OA) Methods The sandwich ELISA method for type Ⅱ collagens was develop,serum levels of type Ⅱ collagens in OA patients and control group were acsayed,and compared with the levels of serum proteoglycans Results There is a high correlation between type Ⅱ collagens and proteoglycans Serum levels of type Ⅱ collagens and proteoglycans were higher in OA patients ( P<0 01,P <0 05) Type Ⅱ collagenlevels were higher ( P< 0 01) and proteoglycanlevels were lower ( P< 0 01) in OA animals X rays showed that type Ⅱ collagenlevels in groups B and C were higher ( P< 0 05) Type Ⅱ collagen decreased ( P<0 05 ) after tibial osteotomy Conclusion Type Ⅱ collagens and proteoglycans can reflect the metabolism of cartilage in OA patients The type Ⅱ collagens are sensitive
