摘  要:目的 :研究无毛小鼠细胞遗传学及生物学特性。方法 :采用血液学常规法、α醋酸萘酯酶染色法(ANAE)、组织HE染色法、骨髓染色体G带方法、骨髓嗜多染红细胞微核法、测交实验法进行细胞遗传学及生物学特性的研究。结果 :①豫医无毛小鼠雌雄间血液学 5项指标均无显著性差异。②豫医无毛小鼠细胞免疫与正常昆明小鼠无异常。③各年龄段无毛小鼠表皮均有一层角化过度的角质层 ,毛干消失 ,毛球结构不正常 ,毛囊被一些角化物质充填。但真皮层差别较大。④豫医无毛小鼠染色体G带无异常 ,并在此基础上绘制了近交系豫医无毛小鼠G带核型模式图。⑤无毛小鼠骨髓嗜多染红细胞对环磷酰胺及煤焦油致突变作用与昆明小鼠相一致 ,是一种评价化学物遗传毒性的有效模型。⑥无毛小鼠无毛性状遗传完全符合孟德尔基因分离规律 ,且和性别无关。结论 :①成功的培育了国内无毛小鼠新品系 ,填补了国内没有无毛小鼠品系的空白。②血液学常规指标的分析、生长发育、免疫能力、皮肤组织结构等一系列生理生化指标的测定 ,首次揭示了豫医无毛小鼠的生物学特性。③首次报道了豫医无毛小鼠染色体G带核型及模式图 ,为突变基因定位奠定了基础。④确定突变基因的类型为纯合单一隐性遗传 ,近交系毛色基因可能为AABBccDD 。Objective: To study the cytogenetics and biology characteristics of Yuyi hairless mice. Methods: By using routine hematological methods, assay of the Lymphocytes labeled with intercellular acid, naphthyl acetute esterase (ANAE), HE stain, the G-handing of the chromosomes methods, the micronuleus of bone marrow polychromatic erythroblasts (PCE) and the coat color genes were tested by mating inbred strain mice with DBA/2, 615 mice carrying the multiple recessive gene respectively.Results: ①Female and male have no signiticant difference in 5 index.②there is no distinctive difference in the cellular immunity between Yuyi hairless mice and wild Kunming mice.③At each age stage, Skin appears over-cornified cuticle, disappeared hair shaft and abnormal hair bulb and hair follide that filled with some cornified substances, but the dermis varied. ④There is not signicant difference on the G banding patterns of the chromosomes between Yuyi hairless mice. ⑤The mutagenlaty of induced by [HT6SS]·6·河南医学研究[HT6SS]第12卷 cyclophosphamide and far Yuyi hairless mouse was same as kunming mice. ⑥The hair shedding character passed to its offspring filled with Mendel's separation law wittion sex-linled. Conclusion: ①the new strain of laboratory animal was developed. There was the hairless mice in China at first.②The biology characteristics of Yuyi hairless mice were reported at first with hematology index, growth and development , immunity index and structure of skin tissue et al, ③The G banding patterns of the chromosomes in Yuyi haieless mice was reported at first.it was used in looking for mutation gene.④The mutation gene of shedding hair is a single recessive gene on autosome, and the genetype were AABBccDD、AaBbccDD in inbred strain and closed colony.⑤ Yuyi hairless mice was a kind of new species experiment animal for evaluation on chemical genetic toxicity.
