摘  要:目的 研究干扰素 (IFN)对培养的大鼠胸主动脉平滑肌细胞内骨桥蛋白表达的影响。方法 大鼠平滑肌细胞被随机分为对照组 ,IFN γ不同浓度 (50 ,50 0 ,1 0 0 0 ,2 0 0 0U ml)及时间 (2 4h ,48h)干预组 ,用RT PCR及Western blotting技术结合光密度扫描分析 ,观察IFN对培养的大鼠胸主动脉平滑肌细胞内骨桥蛋白表达的影响。结果 IFN γ (1 0 0 0U ml)明显刺激平滑肌细胞内骨桥蛋白的表达 ,2 4h干预组RT PCR骨桥蛋白与 β 肌动蛋白mRNA比值 (1 0 34± 0 0 2 9)和 48h干预组比值(1 0 64± 0 0 1 5)分别较对照组提高了 2 4 49% ,2 8 0 3 % ;Western blotting分析 2 4h干预组骨桥蛋白量(1 4 6 65± 5 1 9)较对照组提高 71 1 8% ;48h后 (1 50 49± 1 2 87)较对照组提高 75 66 %。结论 说明IFNObjective To explore whether or not the function of interferon γ(IFN γ) in promoting osteopontin gene expression and protein synthesis of rat vascular smooth muscle cell.Methods Cultured cells were incubated with different concentration of IFN γ for different hours, then RT PCR and Western blotting analysis were used to observe the effects of IFN γ on osteopontin gene expression of rat vascular smooth muscle cells Results IFN γ(1 000 U/ml )could obviously induce gene expression and protein synthesis of osteopontin, RT PCR results showed that the ratio of mRNA of osteopontin and β actin in cells incubated with IFN γ for 24 and 48 hours increased by 24 49%, 28 03% respectively, as compared with that of the control ( P <0 05), and Western blotting analysis also showed that protein synthesis of osteopontin in cells incubated with IFN γ for 24 and 48 hours increased by 71 18%, 75 66% respectively, as compared with that of the control ( P <0 05) Conclusion IFN γ could stimulate osteopontin gene expression and protein synthesis of rat vascular smooth muscle cells
