摘  要:以landersberg生态型的拟南芥为试材,建立了一套简便有效的组培体系。无菌苗的生长采用竖直培养,易于收集外植体。莲座叶、叶柄、下胚轴、根分别作为外植体在B5+5 mg/L2,4-D+0.5 mg/L KT培养基上诱导愈伤组织,结果表明,莲座叶作外植体出愈慢,出愈率低,愈伤组织质量较差;叶柄、下胚轴和根作外植体出愈快,且愈伤组织质量好,后期易分化。根外植体在MS+5 mg/L KT+0.1 mg/L IAA配方的出芽诱导培养基上诱导2~3周后,愈伤组织能有效分化出芽,分化率达100%。分化出的芽移至MS+2 mg/L NAA培养基上竖直培养,2周左右诱导分化出根,然后移至MS培养基上开花结果。A convenient and efficiency system of arabidopsis (landersberg)tissue culture was set up in which the culture of sterile seedling was done vertically for being easy to collect explants. The callus was induced on the medium of B5 + 5 mg/L 2,4 - D + 0. 5 mg/L KT. When different ex-plants were cultured on the callus inducing medium, it was found that callus appeared slowlier from rosettes than from leafstalk, hypocotyl and roots. And that the callus of leafstalk, hypocotyl and roots were better and they were easier to induce shoots than that of rosettes. 2 - 3 weeks-old callus of roots could make differentiation efficiently in shoot inducing medium (MS+ 5 mg/L KT + 0.1 mg/L IAA), with the differentiation rate of 100% . After the differentiated shoots were moved to root inducing medium(MS+ 2 mg/L NAA), roots appeared about 2 weeks later. Then the seedlings were moved to MS growth medium until they flowered and fruited.
