摘 要:本文以中药小檗碱作为分子探针,在0.01mol·L-1醋酸-醋酸钠缓冲体系中,用紫外-可见吸收及荧光光谱法研究了Ru?、Rh?、Pd?三种贵金属离子与DNA的键合相互作用。实验发现Ru?离子对小檗碱-DNA二元体系的荧光有较强的猝灭作用;而Rh?、Pd?两种离子则对该二元体系产生显著的荧光敏化作用。考察了EDTA对贵金属离子、小檗碱及DNA三元混合体系的荧光光谱的影响,初步探讨了贵金属离子与DNA可能的键合机理。Studies on interactions of Ru? , Rh? , Pd? with DNA have been carried out using berberine(BER)as the probe molecule by fluorescence and UV Vis absorption spectroscopy. It was found that the fluorescence intensity of BER DNA system at 526nm was strongly quenched when Ru? was added to the BER DNA system; On the contrary, the fluorescence emission of the system was enhanced when Rh? , Pd? were added to it, respectively. Hyperchromicity, spectral red shifts and isosbestic points were observed in absorption spectra of BER DNA solution in the presence of Ru? , Rh? , Pd? ions. Results show that both Pd? and Rh? ions can bind with DNA to form BER DNA Metal ion complexes in solution, but Ru? ion can bind DNA to replace berberine molecules binding to double helix.
