摘 要:建立了测定动物体液中S型盐酸戊乙奎醚异构体的高效液相色谱法,研究了S型异构体在小鼠体内的排泄。以SpherisorbSiO2色谱柱(150mm×4 6mmi d ,5μm)分离S型异构体,外标法定量。S型盐酸戊乙奎醚异构体的线性范围为0 5~100 0mg/L,最低检测质量浓度为0 1mg/L。肌注给药后不同时间内,S型异构体在小鼠尿中的比值(S 1/S 2)存在显著性差异。48h内各异构体在尿中的排泄量明显不同,说明S型异构体在小鼠体内可能呈现立体选择性排泄。此法简便、经济,可用于动物与人体中S型盐酸戊乙奎醚异构体代谢研究。A high performance liquid chromatographic method was developed for the separation and determination of two (S)isomers of penehyclidine hydrochloride (SPHC: S1, S2) in mouse urine. A Spherisorb SiO2 column (150 mm×46 mm id, 5 μm) was used with a mobile phase consisting of methanolethyl etherammonia water (8∶2∶01 in volume ratio). The flow rate was 08 mL/min and the monitoring wavelength was 254 nm. The calibration curve was linear from 05 to 1000 mg/L of SPHC of mouse urine. The detection limit of SPHC was 01 mg/L for the urine and the recoveries were more than 90% for S1 and S2. The ratio of S1 to S2 in mouse urine within the sampling period was different after dosing. There was an obvious difference for the cumulative excretion of (S)isomers in mouse urine after intramuscular injection of a single dose of SPHC (30 μg). The results suggested that there could be the stereoselective excretion to SPHC isomers in mice. The method is simple and lowcost and can be applied to study the excretion and metabolism of SPHC in animals and human.
