摘  要:建立了测定动物体液中S型盐酸戊乙奎醚异构体的高效液相色谱法,研究了S型异构体在小鼠体内的排泄。以SpherisorbSiO2色谱柱(150mm×4 6mmi d ,5μm)分离S型异构体,外标法定量。S型盐酸戊乙奎醚异构体的线性范围为0 5~100 0mg/L,最低检测质量浓度为0 1mg/L。肌注给药后不同时间内,S型异构体在小鼠尿中的比值(S 1/S 2)存在显著性差异。48h内各异构体在尿中的排泄量明显不同,说明S型异构体在小鼠体内可能呈现立体选择性排泄。此法简便、经济,可用于动物与人体中S型盐酸戊乙奎醚异构体代谢研究。A high performance liquid chromatographic method was developed for the separation and determination of two (S)isomers of penehyclidine hydrochloride (SPHC: S1, S2) in mouse urine. A Spherisorb SiO2 column (150 mm×46 mm id, 5 μm) was used with a mobile phase consisting of methanolethyl etherammonia water (8∶2∶01 in volume ratio). The flow rate was 08 mL/min and the monitoring wavelength was 254 nm. The calibration curve was linear from 05 to 1000 mg/L of SPHC of mouse urine. The detection limit of SPHC was 01 mg/L for the urine and the recoveries were more than 90% for S1 and S2. The ratio of S1 to S2 in mouse urine within the sampling period was different after dosing. There was an obvious difference for the cumulative excretion of (S)isomers in mouse urine after intramuscular injection of a single dose of SPHC (30 μg). The results suggested that there could be the stereoselective excretion to SPHC isomers in mice. The method is simple and lowcost and can be applied to study the excretion and metabolism of SPHC in animals and human.
