摘 要:目的:利用基因芯片技术研究亚砷酸钠对免疫功能相关基因表达的影响 ,探讨砷的分子遗传学机制。方法 :将肝细胞 c DNA克隆库的基因点在芯片上 (每个克隆的基因尚未知 ) ,然后与暴露于不同砷浓度肝细胞中得到的 c DNA第一链杂交 ,对表达有差异的基因进行基因测序 ,再明确基因 ,最后筛选出与免疫功能相关的基因。 结果 :砷可通过抑制细胞角蛋白 8基因 (CK8)和人类白细胞抗原 A基因 (HL A- A)的表达来影响机体免疫功能。 结论 :从基因水平研究砷对免疫功能的作用机制是可行的 。Objective: Effects on sodium arsenite on gene expression contacted with immune function were studied by using gene chip to explore molecular genetic mechanism of NaAsO\-2. Methods: Effects caused by NaAsO\-2 on unkown gene expression through exploring the hepatic cells were dripped on chips and then hybrided with the first strand of cDNA from hepatic cells exposing to different concentrations of NaAsO\-2. Gene sequence of clone differently expressed was detected and defined and to find out those genes which were associated with immune function. Results: NaAsO\-2 related to immune function and inhibited HLA A and CK8. Conclusions: The possible mechanism of NaAsO\-2 effecting on immune function was appraised from gene level and showed its toxical damages to immune system.
