摘 要:目的 :探讨过氧亚硝基阴离子 (ONOO )对体外成骨细胞增殖的影响及丹皮酚 (Pae)对抗ONOO 的作用。方法 :采用改良的淬灭流动反应方法 ,体外制备ONOO 。以不同终浓度加入成骨细胞培养体系中 ,用MTT法观察在不同时相对成骨细胞增殖的影响 ,并用不同终浓度的Pae消除ONOO (10 0 0 μmol/L)对成骨细胞增殖的影响。结果 :低浓度ONOO (5 0~ 2 0 0 μmol/L)对培养的成骨细胞有促增殖作用 ;高浓度ONOO (10 0 0 μmol/L)则明显抑制成骨细胞增殖。Pae(10 - 4~ 10 - 7mol/L)可消除ONOO (10 0 0 μmol/L)对成骨细胞增殖抑制的作用。结论 :低浓度ONOO 促成骨细胞增殖 ,高浓度ONOO 抑制成骨细胞增殖。Pae可消除高浓度ONOO 对成骨细胞增殖的抑制作用。Objective: To investigate the effect of peroxynitrite (ONOO ) on proliferation of osteoblasts in vitro and the antagonizing effect of paeonol on it. Methods: ONOO was produced by modified chemical synthesis with a quenched flow reactor. The inhibitory effect of ONOO on osteoblast proliferation in different time phase was observed with MTT method by adding different final concentration of ONOO into the cultured osteoblast system, and the antagonizing effect of paeonol on the inhibition of ONOO (1000μmol/L) was eliminated by adding paeonol of different concentration into the culture. Results: ONOO in low concentration (50-200μmol/L) could promote the osteoblast proliferation, but it in high concentration (1000μmol/L) inhibited the osteoblast proliferation significantly. Paeonol of concentration with in 10 -4 -10 -7 mol/L could antagonize the inhibition of ONOO on osteoblast proliferation. Conclusion: Low concentration ONOO promotes, but high concentration inhibits the proliferation of osteoblast. Paeonol could eliminate the inhibitory effect of ONOO in high concentration.
