不同浓度珍霉素(17997)滴眼剂对兔实验性单纯疱疹性角膜炎的疗效

摘　　要：目的 :探讨抗病毒药珍霉素 (17997)在兔实验性单纯疱疹性角膜炎中的治疗剂量范围。方法 :观察 5mg·L- 1,10mg·L- 1,2 0mg·L- 1珍霉素滴眼剂 ,0 .1%阿昔洛韦滴眼剂及赋形剂 (随机分成每组10～ 11只眼 ,每日滴眼 5次 ,每次 2滴 ,连续 12d)对兔实验性单纯疱疹性角膜炎的疗效 ,采用双盲法用裂隙灯观察兔角膜病变情况并评分。同时进行兔结膜囊泪液膜病毒分离并计算病毒滴度半数组织细胞感染剂量 (TCID50 )。结果 :各实验浓度的珍霉素滴眼剂对实验性单纯疱疹性角膜炎均有明显疗效 ,可减轻角膜病变程度 (P <0 .0 5 ) ,减少角膜排毒量(P <0 .0 1) ,并与阿昔洛韦滴眼剂比较差异无显著意义。结论 :珍霉素是一种高效的新型抗单纯疱疹性角膜炎的药物 。AIM: To investigate the dosages of antiviral agent 17997 in experimental herpes simplex keratitis(HSK). METHODS: In my study, ten to eleven eyes with herpes simplex keratitis were treated with 5 mg·L -1 , 10 mg·L -1 , 20 mg·L -1 eye drops of 17997 in treatment groups 5 times daily, 2 drops once for 12 d, 0.1 % acyclovir and vehicle eye drops were used in control gorups, and the efficacy was observed. The severity of HSK was double blinded evaluated by slit lamp examinations and scored. Virus sheddings in the tear film and conjunctival sac were isolated and TCID 50 (50 % tissue culture infective doses) were calculated. RESULTS: The results showed a same significant reduction in the severity of HSK in the groups that received 17997 eye drops as 0.1 % acyclovir eye drops ( P <0.05), as well as the mean virus sheddings ( P <0.01). CONCLUSION: 17997 is a new prosperous drug against HSK, with high efficacy.
