摘 要:用PDS-1000/He型基因枪将抗除草剂基因转入水稻的胚性愈伤组织中,获得了转基因水稻植株。通过报告基因gus的瞬间表达研究,优化了基因枪法转化水稻的各种参数。结果表明:DNA金弹到靶细胞的距离为9cm,每皿轰击2次时,愈伤组织的瞬间表达效率高达90%,经X-Gluc染色,每块愈伤组织的蓝点可达100~200个。转基因植株经gus基因的组织化学分析、BASTA除草剂叶片涂布和PCR检测,初步证明外源基因已整合到水稻基因组中,并得以表达。Herbicide resistant gene was delivered into rice (japonica) embryogesis callus induced from mature rice embryo via particle bombardment method. Six transgenic plants were obtained. Parameters affecting transformation efficiency were studied by gus transient expression. When the distance between DNAgold pellet and the target cell is 9 cm, bombarded for 2 times per dish, the transformation efficiency can be as high as 90%, and 100~200 blue spots can be detected on the callus. It was demonstrated that the foreign gene had been inserted into rice genome and expressed in transgenic rice plant by gus biochemical analysis, spraying herbicide on the leaf and PCR analysis.
