摘 要: 用分子沉积自组装技术制备了阳离子化和阴离子化的辣根过氧化物酶自组装膜;并用原子力显微镜(AFM)研究了不同离子化辣根过氧化物酶单层及多层自组装膜的形貌结构与自组装膜的活性变化关系。结果表明:阴离子化的辣根过氧化物酶自组装膜的表面形貌比较粗糙,均方根粗糙度(RMS)及酶分子粒径较大,且组装膜的活性比较大。Horseradish peroxidase monolayer and multilayers were assembled on the surface of the PET using layerbylayer adsorption technique in pH 6.8 and pH7.6 . The different topography of the HRP/polyelectrolyte layer was obtained by AFM in tapping mode. Correlation between the surface topography and the activity of the assembled HRP was investigated. The presented results show that the surface RMS of negatively charged HRP films is larger than that of positively charged HRP films. The activity of negatively charged HRP assembly films is also larger than that of the positively charged HRP film.
