摘  要:目的 研究血管内皮生长因子 (VEGF)基因治疗对大鼠缺血皮瓣生存的影响。方法建立大鼠缺血皮瓣的动物模型 ,采用直接注射法转移 pcD2 /hVEGF12 1真核表达质粒于大鼠缺血皮瓣肉膜层 ,术后 7d ,应用苏木素 伊红 (HE)染色、单光子发射计算机断层摄影 (SPECT)及计算机图像分析软件等方法检测皮下血管密度、皮瓣血供和皮瓣成活率。结果 经VEGF基因治疗组的大鼠缺血皮瓣与对照组相比较具有皮下血管密度增加、血液供应增多和皮瓣成活率显著性增高 (P <0 .0 1)。结论 VEGF基因治疗能够诱导新血管形成、增加血流灌注 ,促进缺血皮瓣的生存。Objective To study the effects of vascu la r endothelial growth factor (VEGF) gene therapy on survival of a ischemic skin f lap in the rats.Methods A randomized skin flap measuring 8 cm×2 cm was elevated onto the back of each Sprague-Dawley rat.pcD2/hVEGF 121 eukaryotic expression plasmid was directly injected into the underlying cu taneous maximus muscle of the ischemic skin flap in the rats.On the 7th day afte r operation,single photon emission computed tomography (SPECT),HE staining,an d computer image analytical software system were used to investigate the blood f low of the skin flap,the subdermic vascular density of each flap,and the survi val rate of the ischemic skin flap.Results The blood supp ly to the flap and the subdermic vascular density were signficantly increased in the treatment group as compared with those in the control group (P<0.01). Compared with the saline control group,the ischemic skin flap receiving pcD2/hV EGF 121 had significant greater tissue viability at the end of 7 days (P <0.01).Conclusion The VEGF gene therapy is able to enh ance the survival of the ischemic skin flap by inducing angiogenesis and increas ing vascular perfusion.
