血管紧张素Ⅱ-1型受体拮抗剂对糖尿病大鼠肾脏细胞凋亡及凋亡相关蛋白的影响

摘　　要：目的　探讨血管紧张素Ⅱ 1型受体拮抗剂 (AT1Ra)对糖尿病肾病肾脏细胞凋亡及凋亡相关蛋白Fas和Fas L表达的影响。方法大鼠腹腔注射STZ诱发糖尿病模型 ,每日灌胃给予AT1Ra缬沙坦(4 0mg kg) ,共 12周。采用原位末端标记法检测肾脏细胞凋亡情况 ,流式细胞术检测肾皮质Fas和Fas L表达。结果　与对照组相比 ,糖尿病组肾小球、肾小管凋亡细胞数明显增多 ,Fas和Fas L的表达增强 ,缬沙坦治疗组较糖尿病组凋亡细胞数减少 ,Fas和Fas L的表达减弱。结论　缬沙坦可能是通过影响凋亡相关蛋白Fas和FasObjective To investigate the effects of angiotesin Ⅱ type Ⅰ receptor antagonist on apoptosis and the expression of Fas and Fas L in the kidney of diabetic rats.Methods STZ (65 mg/kg) induced diabetic male Wistar rats were used in the investigation.Vasartan (40 mg/kg) was delivered daily by gavage from the next day of the induction of diabetes for 12 weeks.Apoptosis was evaluated by means of terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL).Flow cytometry was used to detect the expression of Fas and Fas L.Results Compared with those in the kidneys of control group,apoptotic cells were more in number and the expression of Fas and Fas L was higher in the kidney of diabetic rats (P<0.05).The number of apoptotic cells and the level of expression of Fas and Fas L were reduced in Vasartan treatment group (P<0.05).Conclusion Angiotensin Ⅱ type Ⅰ receptor antagonist Vasartan has some renal protective effects on diabetic nephropathy,partly through inhibiting apoptosis by down regulating Fas and Fas L expression.
