摘 要:目的 :研究运动对 2型糖尿病大鼠脂肪组织中肥胖基因 (obesegene ,obgene)表达水平的影响 ,在基因转录水平上探讨 2型糖尿病大鼠obmRNA表达水平的变化 ,以及运动改变 2型糖尿病大鼠obmRNA表达的机制。方法 :4 0只自发发病的 2型糖尿病模型 :OLETF大鼠分成 4组 ,分别为基础饲料非运动组、基础饲料运动组、高脂饲料非运动组和高脂饲料运动组 ;15只LETO大鼠(与OLETF大鼠同系 ,但不同株 ,系不发病对照组 )用基础饲料饲养 ,随机分为两组 :非运动组和运动组 ,分别进行 9周的运动和高脂饮食干预 ,9周后用实时荧光定量RT -PCR法检测每只大鼠腹膜后脂肪组织obmRNA的表达水平。结果 :基础饲料饲养的OLETF大鼠非运动组和运动组脂肪组织obmRNA的平均拷贝数分别为 4 0 2 4× 10 6 ± 5 4 67× 10 5/ μgRNA和 1 2 5 8× 10 6 ± 3 862× 10 5/ μgRNA ;高脂饲料饲养的OLETF大鼠非运动组和运动组脂肪组织obmRNA的平均拷贝数为 4 832×10 6 ± 1 0 34× 10 6 / μgRNA和 1 5 74× 10 6 ± 6 783× 10 5/ μgRNA ;而基础饲料饲养的LETO大鼠非运动组和运动组脂肪组织obmRNA的平均拷贝数分别为 1 979× 10 6 ± 6 4 77× 10 5/ μgRNA和 1 82 4×10 6 ± 4 0 89× 10 5/ μgRNA。OLETF大鼠脂肪组织obmRNA表达水平运动?Objective To research the effect of exercise on obese gene expression in the adipose tissue in OLETF rat, and to illustrate the mechanism of the change of ob gene expression in NIDDM rats and the mechanism of exercise changing ob gene expression. Methods 40 OLETF rats and 15 LETO rats were randomly allocated to six groups: non-exercise chow-fed OLETF group, exercise chow-fed OLETF group, non-exercise high-fat diet OLETF group, exercise high-fat diet OLETF group, non-exercise chow-fed LETO group and exercise chow-fed LETO group. After 9 weeks of exercise and high fat diet on 40 OLETF rats and 15 LETO rats, the level of ob gene expression was detected by real time fluorogenic quantitative RT-PCR method. Results The mean values of ob mRNA expression of non-exercise and exercise OLETF groups feed by chow were 4 024×10 6±5 467×10 5/μgRNA and 1 258×10 6±3 862×10 5/μgRNA,respectively. As for OLETF rats feed by high-fat diet, the mean value of ob mRNA expression of non-exercise and exercise OLETF groups were 4 832×10 6±1 034×10 6/μgRNA and 1 574×10 6±6 783×10 5/μgRNA. As to LETO rats, the mean values of ob mRNA expression of non-exercise and exercise groups were 1 979×10 6±6 477×10 5/μgRNA and 1 824×10 6±4 089×10 5/μgRNA,respectively. Both levels of ob mRNA expression in exercise OLETF groups were significantly lower than those in non-exercise OLETF groups ( P <0 01), the difference of the level of ob mRNA expression between non-exercise and exercise LETO group was not significant in statistics. Conclusion The increase of ob gene expression in OLETF rats and the decrease of ob gene expression in exercise OLETF groups all are a kind of regulating reaction which can maintain the energy metabolism balance.
