摘　　要：目的:以特定的细胞因子对其进行刺激和诱导,使已经体外分离并扩增的兔骨髓间充质干细胞向软骨细胞谱系分化,为半月板组织工程重建提供种子细胞。方法:将已分离并经体外传代培养扩增的兔骨髓间充质干细胞(mesenchymalstemcells,MSCs)在用碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)和转化生长因子β1(transforminggrowthfactor-β1,TGF-β1)协同刺激后,用逆转录-聚合酶链式反应(reversetranscription-polymerasechainreaction,RT-PCR)的方法从分子水平寻找进入软骨细胞分化谱系的证据。结果:已分离并经体外扩增的兔骨髓MSCs在用bFGF和TGF-β1协同刺激后,用RT-PCR的方法探明有pC-I的编码基因表达,证明其已经进入软骨细胞分化谱系。结论:bFGF和TGF-β1能够刺激MSCs的体外增殖并且使其进入软骨细胞分化谱系,为半月板组织工程重建提供了可靠的自体来源种子细胞。AIM:The in vitro isolated and expanded rabbit bone marrow derived mesenchymal stem cells were induced to differentiate into the chondrocytes lineage with the stimulation of basic fibroblast growth factor(bFGF) and transforming growth factor β1(TGF β1), for meniscus tissue engineering reconstruction.METHODS: After stimulated, evidences were looked for at molecular level to prove whether the MSCs had entered the chondrocyte lineage with reverse transcription polymerase chain reaction(RT PCR) technique.RESULTS:The code gene of precollagen I was detected in the post stimulated MSCs with RT PCR technique, which marked the MSCs'entry of into chondrocyte lineage.CONCLUSION: With the stimulation of bFGF and TGF β1,the in vitro isolated and expanded bone marrow derived MSCs could enter into the chondrocyte lineage,and hence, autogenous seed cells can be provided by this method.
