摘 要:目的探讨白细胞介素_8(IL_8)、肿瘤坏死因子_α(TNF_α)在肾缺血 -再灌注损伤 (IRI)中的作用以及山莨菪碱 (654_2)干预的影响。方法采用大鼠IRI模型 ,动态检测血清和肾组织中IL_8TNF_α含量 ,以及肾功能等指标变化。结果单纯肾缺血、再灌注1h肾组织中IL_8、TNF_α水平明显增高 ,随再灌注时间延长两者逐渐下降至对照组水平 ;血清IL_8在单纯缺血和再灌注1h组呈显著降低 ,随再灌注时间延长 ,逐渐接近对照组水平。IRI各组肾系数、血清SUN和Scr水平均高于对照组 ;654_2干预可使肾IRI4h组血清SUN、Scr及肾系数明显高于未干预组。结论IL_8、TNF_α可能在早期肾IRI中起重要作用。Objective To explore the effect of interleukin_8(IL_8)? tumor necrosis factor_α(TNF_α) on renal ischemia_reperfusion injury(IRI) and the effect of 654_2.Methods Using a rat model of renal IRI, the changes in levels of IL_8?TNF_α in renal tissues and serum,and renal function indexes were dynamically observed.Results The levels of IL_8?TNF_α in renal tissues were increased in pure ischemia and ischemia_reperfusion 1 hour(IR1h) group and both decreased to level of controls acompanying prolonged reperfusion. The levels of IL_8 in serum were decreased in pure ischemia and IR1h group and it reached level of controls by prolonging of reperfusion. The levels of serum urea nitrogen(SUN)?serum creatinie(Scr) and renal coefficients were increased to controls in every group of ischemia_reperfusion. In IR4h group,levels of SUN?Scr and renal coefficicients in 654_2 interference group were higher than those in non 654_2 interference group.Conclusions IL_8?TNF_α might play an important role in early renal IRI.654_2 interference might become unfavourable effect in early stage of renal IRI .
