摘 要:[目的 ]探讨乙醇对大鼠神经行为效应及脑组织中相关酶的影响。 [方法 ]体重 160g左右的雄性SD大鼠2 5只 ,随机分为三组 ,每组为 9、8、8只 ,分别以 0 % (对照组 )、2 5 64 % (低剂量组 )、5 1 2 8% (高剂量组 )乙醇溶液 (浓度以V/V表示 )按 1 0ml/10 0g体重一次性灌胃染毒。 3 0min后 ,进行自发活动、转棒实验和方位水迷宫测试。在染毒 1h后 ,断头处死 ,取血测血中乙醇浓度 (BAC) ,取脑组织测丙二醛 (MDA)、超氧化物歧化酶 (SOD)、巯基化合物 ( SH )和胆碱酯酶活性。 [结果 ]高剂量组自发活动中的行走次数显著高于低剂量组和对照组 ;转棒测试 ,高、低剂量组在棒上停留时间均显著低于对照组 (P <0 0 1) ;水迷宫试验学习次数 ,在染毒前对照组明显高于染毒组 ,而在染毒后各组间差异无统计学意义 ;且上述改变与BAC相关显著。高剂量组SOD活性显著低于低剂量组和对照组。丙二醛、巯基水平及胆碱酯酶活性各组间无明显差异。 [结论 ]乙醇可降低大鼠对新环境的适应性、肌肉运动协调能力下降和学习记忆能力的减退 。To explore the effects on neurobehavioral function and neurobiochemical changes induced by ethanol in rats. 25 male Sprague Dawley rats weighed about 160 g were randomly divided into 3 groups. Each group was once administered intragastrically with ethanol solution(0%,25 64%,51 28%,V/V). The intragastrical volume was 1 0 ml/100 g b.w.. Behavioral function including Locomotor Activity,Rotarod and Spatial Water Maze,they were tested at one week before and 30 minutes after exposure to ethanol. Rats were sacrificed by decapitation in one hour after ethanol administration. The blood was sampled and the brains were rapidly removed. The following indicators were assayed:1)Blood alcohol concentration(BAC). 2)Levels of lipid peroxide,glutathione and activity of superoxide dismutase(SOD). The BAC in the high dose group,low dose group and control group were 67 50 mM,63 15 mM,2 88 mM,respectively. The scores of ambulation of locomotor activity increased in the high dose group. The sustaining time of rotarod in the high and low dose groups were shorter than that in the control group. The learning numbers of Spatial Water Maze in the control group were higher than those in the exposure groups before exposure,but there was no statistical difference among the control and exposure groups after exposure. Furthermore,the behavioral alternations above in rats were correlated with BAC. The activity of SOD was significantly decreased in higher exposure group. [Conclusion] The ethanol could diminish the adaptation to new surrounding,impair the abilities of motor coordination,learning and memory in rats. There was a correlation between the alterations mentioned above and the SOD activity of the brain.
