摘  要:目的 研究羟基积雪草甙 (madecassoside ,Md)对人皮肤成纤维细胞 (humanskinfibrob last,HSFb)的增殖及胶原蛋白合成的影响。方法 XTT法研究Md对HSFb增殖的影响 ;放射免疫的方法测定实验组和空白对照组细胞上清液中Ⅰ型和Ⅲ型前胶原氨基端肽原 (procollagenⅠandⅢofaminoterminalpropeptide ,PINP ,PⅢNP)的含量。 结果  (1 )Md在浓度高达 2 0 0 μg/ml时仍未见任何细胞毒性。 (2 )与空白对照组相比 ,Md对HSFb有明显促进增殖作用 (P <0 0 5) ,有效刺激HSFbPINP和PⅢNP的分泌。 (3)Md刺激靶细胞PINP和PⅢNP分泌的生物学作用呈现明显的量效关系。结论 Md在刺激体外培养HSFb增殖及Ⅰ。Objective To study the effects of madecassoside on human skin fibroblasts proliferation and types Ⅰ and Ⅲ collagen synthesis in vitro.Methods The influence of madecassoside on proliferation of cultured fibroblast was determined by XTT method,and the protein content of PINP and PⅢNP in the culture medium of treated groups and control groups was determined by radioimmunoassay.Results (1)Madecassoside had no cytotoxicity at the concentration of 200μg/ml.(2)Compared with control groups,the increases of cells proliferation and an obvious stimulation of PⅠNP and PⅢNP protein secretion were observed in the fibroblast treated with madecassoside ( P< 0 05).(3)The pharmacological action of madecassoside exhibited a dose dependent manner.Conclusion Madecassoside has significant biological activities on stimulation of the cells proliferation and the type Ⅰ and Ⅲ collagen synthesis in cultured fibroblasts in vitro.
