丙型肝炎病毒5’-非编码区与NS 5b区酶切分型对比研究

摘　　要：探讨北京与河南地区丙型肝炎病毒(HCV)基因型分布情况以及HCV 5’-NCR和HCV NS5b两区域分型结果之间的差异。对56例慢性丙型肝炎和80例有偿供血员的HCV RNA阳性血清。同时进行HCV 5’-NCR和HCV NS5b酶切分型研究。HCV 5’-NCR和HCV NS5b两区域的Ⅱ/1b型感染率北京地区分别占85.7％和87.5％,河南地区分别占68.8％和70.0％。HCV Ⅱ/lb型为北京和河南地区HCV感染的优势株,同时HCV 5’-NCR和HCV NS5b区酶切分型结果间具有良好的相互对应关系。To investigate the genotypes of HCV infection in Beijing areas and in Henan areas, the differences of HCV genome with restriction endonuclease between 5' - noncoding region (5' - NCR) and nonstructural gene 5b(NS5b) region were compared. RT - PCR for 5' - NCR and NS5b region were used in the same time to detect HCV RNA in 56 chronic hepatitis C patients and 80 blood donors, and the positive PCR products were typed by restriction endonuclease. The results showed that 48(85.7%) at5' -NCR and 49(87.5%) at NS5b region were HCV type Ⅱ/1b infection in Beijing areas, 55(68.8%) at 5' - NCR and 56 (70.0% ) at NS5b region were HCV type Ⅱ /1b infection in Henan areas. These results indicated that HCV infection in Beijing or in Henan areas were mainly type Ⅱ/1b, and the HCV genome with restriction endonuclease at NS5b region were in accordance with that at 5' -NCR.
