摘  要:目的 比较成纤维细胞 (FB)在猪无细胞真皮基质 (ADM)上的黏附及生长特性 ,探讨 ADM作为组织工程化皮肤支架材料的可行性。 方法  FB2 ml细胞浓度为 2 .5× 10 5/ ml,分别接种于经戊二醛交联、未经戊二醛交联、戊二醛交联不含基底膜和戊二醛交联并打孔形成网状的猪 ADM中孵育 ,相同数量 FB接种于空白培养皿中作为对照 ,接种后 1、3及 5天行细胞计数 ,黏附有 FB的 ADM行 HE染色观察。 结果  FB在经戊二醛交联、未经打孔完整的ADM上的黏附增殖优于未经戊二醛交联或戊二醛交联并打孔的 ADM,是否含完整基底膜对成纤维细胞黏附增殖无明显影响。 结论 采用经戊二醛交联、完整的 ADM有利于 FB的体外培养 ,ADM经一定的修饰后可能作为组织工程化皮肤的支架材料。Objective To compare the attachment and growth of fibroblasts on the different porcine accellular dermal matrix (ADM) so as to find the suitable scaffold for tissue engineering skin. Methods Fibroblasts (5×10 5) were seeded on 4 kinds of ADMs which were crosslinked with glutaraldehyde, uncrosslinked, crosslinked with glutaraldehyde and removed basement membrane, corsslinked with glutaraldehyde and then meshed. The same density fibroblasts were seeded on petri dish as a control. Cell count was done on the 1st, 3rd, 5th days after seeding. The attachment of fibroblasts on ADMs was observed by HE staining. Results The growth and attachment of fibroblasts on crosslinked and non meshed ADM increased markedly when compared with the others. There was no obvious difference between the groups of with or without basement membrane. Conclusion The above results indicate that non meshed and corsslinked with glutaraldehyde ADM is more suitable for the attachment and growth of fibroblasts than the others and that the modified ADM can be used for the scaffold of tissue engineering skin.
