摘  要:栽培甜菜与白花甜菜杂交而获得的单体附加系M14,其传递率在95%以上,单体附加系的传递率如此高的原因可能是因为M14品种中有无融合生殖基因存在。为了寻找M14品种中无融合生殖基因,本试验在花期分别对二倍体甜菜A2Y和M14品种的花器进行取样,对它们的总RNA进行逆转录,RT-PCR反应,并对其中各种反应参数进行筛选和优化,最终建立起对它们进行mRNA差异反应的最佳体系。此体系的建立为寻找二倍体甜菜A2Y和M14品种之间的差异条带提供了一个稳定、方便的方法。同时也为以后进一步寻找到无融合生殖基因,做了一项基础而又有价值的前期工作。mono-somic additional line M 14 was hybrid between cultivated sugar beet and B.corolliflora zoss,and its transfer rate is more than95%,apomixes gene in the M 14 variety is the reason that the transfer rate of mono-some additional line is so high.In order to find the apomixes gene in the M 14 variety,the flora organ of diploid A 2Y and M 14 variety was sampled respectively at the flowering phase,converse transition and RT-PCR reaction was carried out for their total RNA,each reaction coefficient was boltinged and improved in the experiment,at last the best system of mRNA difference reaction was founded.The foundation of the system provided a stability and convenient method for finding difference lace between diploid sugar beet A 2Y and M 14 variety.In the same time it is a basic and valuable ex-period work for further find out the apomixes gene.
