助孕3号方及拆方防治肾虚黄体抑制动物流产模型的实验研究

摘　　要：目的:了解助孕3号方及拆方对肾虚黄体抑制动物流产模型的治疗作用及机理。方法:对妊娠大鼠以羟基脲和米非司酮建立肾虚黄体抑制病证动物流产模型,分别用助孕3号方、补肾方、健脾方中药水提液及黄体酮治疗,同时设立未用药物治疗的模型对照组和正常妊娠组。测定各组流产率、子宫卵巢组织形态学变化、血清P及子宫蜕膜PR的含量。结果:补肾健脾的助孕3号方和补肾方均可增强黄体功能,提高血清孕酮含量,增加子宫蜕膜孕激素受体mRNA的表达,从而降低肾虚黄体抑制动物流产模型的流产率,单纯健脾方无此作用。结论:补肾药在防治黄体功能不全引起的流产方面起主导作用。Objective: To examine the therapuetic effect and the mechanism of the Traditional Chinese Medicine 'prescription Zhuyun Ⅲ 'and its ingredings'prescription Bushen'and 'prescription Jianpi' on spontaneous abortion (SA). Methods: An aborting kindney-deficiency rat model was created by giving hudroxyurea, and then luteal phase defect model was created by giving Mifepristone. Fourty-five pregnant SD rats were randomly divided into 6 groups. Group A was given Hydroxyurea+Mifepristone+Prescription Zhuyun Ⅲ; Group B was given Hydroxyurea+Mifepristone+Prescription Bushen;Group C was given Hydroxyurea + Mifepristone + Prescription Yianpi; Group D was given Hydroxyurea + Mifepristone + Progesteron injec-tionjGroup E was given Hydroxyurea+Mifepristone+NS;Group F was serve as normal pregnant control group. The indexes of weight, abortion rate, serum progesteron level, the positivtity rate of decidua progesteron receptor (PR) and the pathological changes of uterus and ovary were observed in different groups. Results: 'Prescription Zhuyun Ⅲ ' and 'Prescription Bushen' could strengthen the function of corpus luteum, increase the serum level of progesteron and raise the content of progesteron receptor in decidua. But the 'Prescription Jianpi' whose main functio was strengthening the spleen,did not have the same effect as above. Conclusion: Herbs which could supplement the kindney played a major role in treating the SA which was caused by defect of corpus luteum.
