大鼠肾上腺髓质增生模型的动态观察

摘　　要：目的　探讨利血平建立大鼠肾上腺髓质增生 (AMH)模型的最适剂量 ,观察其长期稳定性。方法　雄性Wistar大鼠 72只 ,随机分为A、B、C、D 4组各 18只 ,A组皮下注射利血平 0 .2mg·kg-1·d-1和 0 .9%生理盐水 0 .6mg·kg-1·d-1,B组皮下注射利血平 0 .4mg·kg-1·d-1和 0 .9%生理盐水 0 .4mg·kg-1·d-1,C组皮下注射利血平 0 .8mg·kg-1·d-1,D组皮下注射 0 .9%生理盐水0 .8mg·kg-1·d-1,每日 1次 ,持续 2个月后停药。分别于停药后 2周、2个月、6个月时测大鼠 2 4小时尿 3 甲氧基 4 羟基苦杏仁酸 (VMA )含量及血儿茶酚胺 (CA)水平 ,然后处死大鼠 ,取出肾上腺 ,作病理切片 ,测肾上腺髓质百分数 ,及AMH阳性率。结果　在各停药时段 ,B、C组与D组相比2 4h尿VMA含量、血浆CA水平和髓质百分数均有显著性升高 (P <0 .0 5 ) ,这三组各值与停药 2周相比差异均无显著性 (P >0 .0 5 ) ;在停药 6个月时 ,A组各值与停药 2周时相比有显著性下降( P <0 .0 5 ) ,与D组相比差异均无显著性 ( P >0 .0 5 )。结论　利血平所致大鼠AMH模型各生化指标稳定 ,但C组死亡率较高。Objective To investigate the optimal dose of reserpin for establishing a rat experimental model of adrenal medulla hyperplasia (AMH).Methods 72 male Wistar rats were randomly divided into 4 groups.Group A were subcutaneously (sc) injected with reserpin (0.2 mg·kg -1·d -1) and 0.9% sodium chloride (0.6 mg·kg -1·d -1),group B sc with reserpin (0.4 mg·kg -1·d -1)and 0.9% sodium chloride (0.4 mg·kg -1·d -1),group C sc with reserpin (0.8 mg·kg -1·d -1) and group D sc with 0.9% sodium chloride (0.8 mg·kg -1·d -1).3-methoxy-4-hydroxyvanillymandeic acid (VMA) content in the urine and catecholamine (CA) level in plasma were determined at 2nd week,2nd month,6th month after injection.Pathologic studies of adrenal were done.Results At different time points,all values in groups B and C were higher than those in the group D (P<0.05).At 6th month after injection,all values in groups B,C and D were not different from those at 2nd week after injection (P>0.05).The values of VMA and CA in group A were not different from those in group D at 6th month after injection (P> 0.05),but lower than those at 2nd week after injection (P<0.05).Conclusion The rat experimental model of AMH in groups B and C is more steady than group A,but mortality of group C is high.
