应用大鼠拟皮肤模型评价巯基乙酸的皮肤毒作用

摘　　要：[目的 ]探讨巯基乙酸 (TGA)的皮肤毒作用及其拟皮肤模型替代正常皮肤应用于皮肤毒理学研究的可行性。[方法 ]应用导电聚吡咯膜技术扩增大鼠角质形成细胞和成纤维细胞 ,将胶原蛋白与成纤维细胞混合培养构建拟真皮 ;种植角质形成细胞在拟真皮上 ,经培养形成拟皮肤。在拟皮肤和新生大鼠皮肤上涂布巯基乙酸溶液局部染毒 1h ,培养一定时间后对表皮进行组织学观察。 [结果 ]在光镜下可见所构建的大鼠拟皮肤模型具有与正常新生大鼠皮肤相类似的结构 ,包括真皮 ,完整的基底层、分化细胞层与角质层。用 1 %～ 8%浓度的TGA进行 1h染毒后即能引起拟皮肤和新生大鼠皮肤一定程度的病理学改变 ,包括角质层松散、细胞分层模糊、颗粒层消失、基底细胞变性等。两者对TGA的反应有着较大相似性。 [结论 ]使用低于我国化妆品卫生规范 (卫法监发 [2 0 0 2 ]第 2 2 9号 )的浓度 ,TGA接触皮肤 1h即可出现明显皮肤损害的病理学改变 ;大鼠拟皮肤模型对外来毒物损伤可做出类似大鼠皮肤的损伤反应 。To explore the cutaneous toxicity of thioglycolic acid(TGA) and to confirm the possibility of the rat pseudo skin as suitable replacement of rat skin to study cutaneous toxicology. Large number of rat keratinocytes and fibroblasts were obtained by the conductive polypyrrole electrical stimulation technique. The pseudo dermis was formed with a mixture of fibroblasts and type Ⅰ calf collagen,and then the keratinocytes were seeded on it. After a period of 19d submerged culture and air liquid interface culture,a piece of rat pseudo skin was formed. The rat pseudo skin and new born rat skin were topically exposed to TGA for an hour and the pathological changes of them were observed under microscope. It was shown that the rat pseudo skin was similar in morphology to that of newborn rat skin with dermis,basal layer,granular layer and stratum corneum. Topical application of TGA could cause some pathological damages to both rat pseudo skin and rat skin,such as lysis of stratum corneum,acanthosis,disappearance of stratum granulosuma,the vacuolating degeneration of the upper epidermis cells and liquefaction of basal cells and so on. [Conclusion] Obvious damage in both rat skin and rat pseudo skin occurred with topical application of lower dose of TGA than that of National Cosmetics Hygiene Standard. The rat pesudo skin established in this study can be used in cutaneous toxicity study instead of normal skin in some toxicological fields.
