温度和酸碱度对黄精凝集素Ⅱ生物学活性与构象的影响研究

摘　　要：采用圆二色谱和荧光光谱 ,研究了黄精凝集素Ⅱ (PCLⅡ )在不同温度及不同pH值时 ,其活性和分子构象变化的关系 ,结果表明 :PCLⅡ具有较强的温度和酸碱度的耐受性 ,但在80℃ ,pH 2 ,pH 4和 pH 12时 ,其分子构象发生较大变化 。Circular dichroism(CD) and fluorescence spectra were used to elucidate the relationship between the activity and molecular conformation of Polygonatum cyrtonema Hua.,Lectin Ⅱ(PCLⅡ) under different temperature and pH Value. The results showed that PCLⅡ was stable in various temperature and pH between pH 4～9.5 and below 80℃, but the molecular structure and biological activity changed much under pH 2,pH 4,pH 12 and 80℃, and the activity of PCLⅡ became weak.
