摘 要:目的 探讨颗粒性脑蛋白聚糖 (PG)与脑发育和老化的关系。方法 纯种SD大鼠随机分为 3组 :初生组、青年组和近老年组。脑组织经脱脂处理后 ,4mol·L-1盐酸胍提取各组大鼠颗粒性脑PG ,DEAE Sephacel柱层析分离纯化 ,Sepharose 4B柱层析分析其分子大小 ,双向电泳法分析其糖胺多糖 (GAG)的组成。 结果 颗粒性脑PG的提取率随增龄而降低 ;大分子硫酸软骨素蛋白聚糖 (CSPG)主要存在于初生组和青年组 ;小分子硫酸皮肤素蛋白聚糖 (DSPG)的比例随增龄而增高 ,而小分子CSPG的比例则随增龄而降低。结论 颗粒性脑PG随动物年龄的增长发生一定的变化 ,与脑的发育和老化密切相关。Objective To study the association of particulate brain proteoglycan (PG) with development and aging of the brain. Methods SD rats were divided into three groups: new born, young adult and near old groups. After brain tissues were defatted, particulate brain PGs of each group were extracted with 4?mol·L -1 of guanidine HCl and purified by DEAE sephacel chromatography, their molecular sizes were identified by Sepharose 4B chromatography and their glycosaminoglycan(GAG) compositions were characterized by two dimensional electrophoresis. Results The extraction rates of particulate brain PGs decreased with age; the large molecular size particulate CSPG mainly existed in new born and young adult groups; the percentage of small molecular size DSPG increased and CSPG declined with age. Conclusion Particulate brain PGs change with age and are related to brain development and aging.
