摘  要:目的 研究人参皂苷Rg3(Ginsenoside Rg3)对体外培养视网膜色素上皮 (RPE)细胞增生的直接抑制作用及可能机制。方法 通过活细胞计数法与MTT比色法分别检测不同浓度 (10、2 0、5 0、80、10 0、15 0mg L)Rg3和Rg380mg L在不同作用时间 (6~ 12 0h)对RPE细胞增生及代谢的影响。结果 Rg3组细胞增殖受到抑制并出现细胞脱落 ,Rg310 0mg L具有最强的抑制效应 ,其明显抑制效应在 6h即出现 ,96h达高峰。Rg3组A值明显低于对照组 ,RPE细胞生存率下降。结论 Rg3可能通过抑制钙内流促进钾外流改变细胞膜电位、干扰RPE细胞代谢 。Objective To investigate the effects of ginsenoside Rg 3 on proliferation of cultured retinal pigment epithelium(RPE) cells and the probable mechanisms.Methods Cultured RPE cells were treated with Rg 3 by various concentrations(10,20,50,80,100,150mg/L) and with Rg 3 80mg/L at different times(6~120h),cells proliferation and metabolism were evaluated by cell count method and MTT colorimetric assay.Results There was inhibition on proliferation of RPE cells and some cells detached from the wall in Rg 3 group.The best concentration of Rg 3 was 100mg/L.The significant inhibitory effect of Rg 3 occurred at 6h and the strongest inhibition of Rg 3 occurred at 96h.The A value in Rg 3 groups was significantly lower than that in control group and the survival rate of RPE cells lowered.Conclusion The results suggest that Rg 3 could inhibit the proliferation of RPE cells in a dose dependent and time dependent manner,the probable mechanism is that Rg 3 could cause an inhibition of Ca 2+ influx and stimulation of K + efflux,so the membrane potential changes and the mebabolism of cell is disturb.
