腹膜透析患者合并腹膜炎症对腹腔巨噬细胞hsp70、iNOS基因表达的影响

摘　　要：目的 :探讨腹膜炎症对腹膜透析患者腹腔巨噬细胞hsp70、iNOS基因表达的影响。方法 :应用原位杂交、免疫组化及斑点印迹技术对试验组和对照两组腹腔提取液巨噬细胞iNOSmRNA、iNOS及hsp70进行测定。结果 :斑点印迹结果显示胞内iNOSmRNA、iNOS及hsp70的信号强度 ,实验组第 1天大于实验组第 5天及对照组 ,差异显著性 (P <0 0 1)。腹膜炎症初期 ,iNOSmRNA、iNOS及hsp70均为高表达。结论 :腹膜透析伴腹膜炎患者腹腔巨噬细胞hsp70、iNOS基因高表达 ,其产物NO、hsp70在腹腔局部免疫中有重要意义。Objective: To investigate the effect of peritonitis on hsp 70 and iNOS gene expression of peritoneal macrophages in peritoneal dialysis patient and to examine the immunological merit elicited by hsp70 and NO. Methods: By situ hybridization, immunohistochemistry, dot blot techniques and chromatogram scanning, the contents of iNOSmRNA, iNOS and hsp70 of peritoneal macrophages were determined in both control group and test group. Results: As seen from the scanning intensities of dot blot signals,the contents of iNOS and hsp70 on day l of test group were significantly higher than that on day 5 of test group and on day 1 and day 5 of control group ( P <0.01). Conclusion: Among peritoneal dialysis patients with peritonitis, hsp70 and iNOS genes in peritoneal macrophage show higher expression at the initial stage of peritonitis and expression products NO and hsp70 play an important role in local immunity of peritoneal cavity.
